Sarja kertoo Marvelin fiktiivisen S.H.I.E.L.D. (Strategic Homeland Intervention, Enforcement and Logistics Division) järjestön toiminnasta. Agentti Phil Coulson kutsuu koolle pienen, valikoidun agenttitiimin, jonka tehtävänä on tutkia outoja ja epänormaaleja tapahtumia, sekä yliluonnollisia kykyjä omaavia henkilöitä. Coulsonin agentit ovat itsekkin enemmän tai vähemmän supersankareita, jotkut ovat toimineet kentällä jo pitkään, osa värvätään toimiston pöydän takaa. Ryhmään saadaan myös brittiläiset tiedemiehet Leo Fitz ja Jemma Simmons. Coulson värvää mukaan vielä tietokonespesialisti/hakkeri Skyen - naisen, jolla ei ole nimeä eikä menneisyyttä.
