Gangs of London lähtee käyntiin siitä sattumanvaraisesta hetkestä, kun Lontoon vaikutusvaltaisimman rikollisryhmän patriarkka Finn Wallace tapetaan. Wallacen kaksi poikaa ja leski päättävät selvittää murhaajan ja laittaa tämän vastuuseen. Koston edetessä perheen ote muista alamaailman ryhmittymistä alkaa kuitenkin livetä. Lontoon alamaailman valtatyhjiötä täyttämään lähtevät lukuisat kansainväliset ryhmittymät, joista jokainen haluaa kasvattaa omaa siivuaan vallasta.
